چکیده

The pH profiles of the apparent biomolecular rate constants for the reaction f free EO with [2-'H]and [22H]phenylalanine (evaluated as (kr)H and (kr)H*, respectively, from rapid kinetic measurements and as the equivalent steady state coefficients ( & l ) ~ and (+I-')"*) show that k , = k', k2 >> k-' << k2* and that the ionization controlled by K', (= kL,/k,) is at virtual equilibrium. The expression for (kr)H is, therefore,

Nitroalkanes as reductive substrates for flavoprotein oxidases.

Nitroalkanes have been found to be general reductive substrates for D-amino acid oxidase, glucose oxidase and L-amino acid oxidase. These enzymes show different specificities for the structure of the nitroalkane substrate. The stoichiometry of the D-amino acid oxidase reaction is straightforward, consisting of the production of one mole each of aldehyde, nitrite and hydrogen peroxide for each m...

Nucleophilic addition reactions of free and enzyme-bound deazaflavin.

DeazaFMN-containing glycolate oxidase has been prepared and shown to catalyze the stereospecific transfer of the alpha-hydrogen from substrate to enzyme-bound deazaFMN. The reaction of sulfite, cyanide, and hydroxylamine with several deazaflavin-containing enzymes (glycolate oxidase, D-amino acid oxidase, glucose oxidase, N-methylglutamate synthetase) and free deazaFMN has been examined. All th...

pH and kinetic isotope effects in d-amino acid oxidase catalysis.

The effects of pH, solvent isotope, and primary isotope replacement on substrate dehydrogenation by Rhodotorula gracilis d-amino acid oxidase were investigated. The rate constant for enzyme-FAD reduction by d-alanine increases approximately fourfold with pH, reflecting apparent pKa values of approximately 6 and approximately 8, and reaches plateaus at high and low pH. Such profiles are observed...

Proton release during the reductive half-reaction of D-amino acid oxidase.

Changes in the net protonation of D-amino acid oxidase during binding of competitive inhibitors and during reduction by amino acids have been monitored using phenol red as a pH indicator. At pH 8.0, no uptake or release of protons from solution occurs upon binding the inhibitors benzoate, anthranilate, picolinate, or L-leucine. The Kd values for both picolinate and anthranilate were determined ...

Oxidation-reduction potentials of D-amino acid oxidase.

This paper reports a study of the oxidation-reduction equilibrium of o-amino acid oxidase, a flavoprotein con. taining FAD. The oxidation-reduction potential at 50% oxidation (E;) is -0.004 volt at pH 7.0 and 20”, and therefore about 180 mv higher than that of the free coenzyme (FAD). This difference in oxidation-reduction potential may be described in terms of relative affiity of the apoenzyme...
